Yоu аre interested in studying the secretiоn оf interleukin 21 (IL-21) in the context of Cаndidа albicans Specifically, human peripheral blood mononuclear cells (PBMCs) were isolated from the blood of healthy donors and the cells were stimulated with Candida albicans extract (CAe). The concentration of IL- 21 in the supernatant and number of IL-21 secreting cells were assessed via ELISA and ELISpot, respectively. After looking at the results, the number of IL-21 secreting cells could be detected via ELISpot but the concentration of IL-21 in the supernatant was negligible when measured via ELISA. You decided to repeat the same experiment but this time, when stimulating the cells with CAe, you decide to simultaneously add an antibody that blocks the IL-21 receptor on the PBMCs. Importantly, addition of the anti-IL-21 receptor antibody resulted in both, a measurable IL-21 concentration in the supernatant via ELISA and a quantifiable number of IL-21 secreting cells via ELISpot. Can you explain why adding the anti-IL-21 receptor antibody resulted in a quantifiable ELISA? (Huang et al. Journal of Immunological Methods. 2015. 417:60-66)
