Darrell seems to be a little weaker than he should be at that age, although the other kids seem fine.  The reported family history is negative for any muscle disorders.  Darrell’s saliva sample is sent for DNA testing of the DMD gene (encoding dystrophin) to screen for Duchenne muscular dystrophy (an X-linked recessive disorder). The result comes back likely negative - only a silent (synonymous) substitution was identified. A year later, Darrell’s weakness is a little worse and Isaac seems to be starting to show signs.  Based on this scenario, answer the questions below.  QUESTIONS: If this really is Duchenne muscular dystrophy, how could it be caused by a silent substitution point change? You are suspicious that this point substitution is really just a neutral polymorphism. What evidence would you search for, to test that hypothesis? If this is actually an autosomal dominant condition, how could you explain why neither parent is affected? If the point change is proven to be neutral, and whole genome sequencing finds no candidate DNA sequence variants in the affected kids, what else could be responsible for these symptoms (in general)? Suppose by the next year, the mother starts to show signs of similar weakness, and hears that her first cousin (son of her mom’s sister) is also having some weakness.
