Blооd typing is аccоmplished by аdding а patient's erythrocytes to known antisera and looking for agglutination. Agglutination occurs when specific antigens come in contact with specific antibodies, causing the clumping of the erythrocytes (note, this is very different from clotting). Each erythrocyte has specific markers or antigens that coat the outside of their cells. When these cells come in contact with a specific antibody then agglutination occurs. Blood typing is important since each person carries antibodies to any markers not present on their erythrocytes. Therefore, if an incorrect blood type is given in a transfusion, the antibodies within the plasma of the patient will cause the destruction of the foreign blood cells. To type blood, mix a drop of blood with each of the following antisera: Anti-A, Anti-B and Anti-D. Anti-D is testing for the Rh antigen. Observe each of the wells for agglutination (looks like granules or bits of sand). In the example below, you see agglutination in the B well (meaning they have the B antigen on their erythrocytes) and agglutination in the Rh well (meaning they are positive for the Rh antigen).
