Bаckgrоund: Thyrоxine (T4) is а hоrmone used by vertebrаte animals to regulate the metabolism of all cells. The production of T4 depends on the concentrations of two other hormones, thyrotropin-releasing hormone, and thyroid-stimulating hormone. Thyrotropin-releasing hormone (TRH) comes from a part of the brain called the hypothalamus. This hormone binds to cells of the pituitary gland, which causes these cells to release thyroid-stimulating hormone (TSH). TSH binds to cells of the thyroid gland, which causes these cells to release T4. Being mostly nonpolar, T4 can diffuse through the phospholipid bilayer of any cell, including cells of the hypothalamus and the pituitary gland.   The production of T4 depends on the activity of an enzyme called thyroid peroxidase (TPO) and its substrate, iodide (I-).
