During DNA synthesis there are a number of components (enzymes, nucleic acid sequences, nucleotides, binding proteins, etc.) necessary to separate DNA strands, prevent them from reconnecting, starting, and continuing synthesis of new, complementary strands of DNA. Polymerase Chain Reactions (PCR) is an in vitro (not inside cells) method that allows researchers to synthesize specific sequences of DNA in the lab. The process of PCR consists of several steps of heating and cooling a tube containing DNA: heating it to approximately 95°C to separate the strands, cooling it to approximately 65°C to encourage primer sequences to attach to specific DNA sequences, and then heating it to approximately 70°C for the DNA synthesis machinery to begin building the new DNA strands.
